That's not going to work. RDIMMs are not supported in LGA115x systems.

Supermicro Micro ATX DDR4 LGA 1151 Motherboards X11SSL-F-O

Consider X11SSM-F which supports 8 Sata drives not 6

Cucial 8GB Single DDR4 2133 MT/s (PC4-2133) CL15 SR x4 ECC Registered DIMM 288-Pin Server Memory CT8G4RFS4213

Consider getting a 16GB stick, will mean you can continue adding 16GB sticks to get to maximum capacity over time (Ie 16x4)

SanDisk Ultra Fit™ CZ43 16GB USB 3.0 Low-Profile Flash Drive Up To 130MB/s Read- SDCZ43-016G-G46

Consider getting two so you can mirror boot drive

Seasonic SSR-360GP 360W 80PLUS Gold ATX12V Power Supply

Depends on how many drives you want to end up with.

Fractal Design Node 804 No Power Supply MicroATX Cube Case FD-CA-NODE-804-BL, Black

Western Digital WD40EFRX 4 TB WD Red, 3.5 inch, SATA III 5400 RPM 64 MB Cache Bulk/OEM NAS Hard Drive

How many?

Intel BX80662I36100 3.70 GHz Core i3-6100 3M Cache Processor

Probably. Try it and check your temps while running mprime. If you need better cooling then buy it, but don't until you've tested.

2x4tb drives. I may want to add 2 more on a separate mirror. I did not think of 2usbs drives, that really good. I always wondered if 1x16gb vs 2x8gb sticks made any difference.

2x16GB is even better. You can now use ZFS to mirror your boot drives, and since USB sticks have a habit of failing this is very good advise.

Please consider just using an inexpensive SSD as a boot drive. USB sticks are mostly to save SATA ports, but that's not a problem here. It's possible to mirror these also, if desired.

If I save snapshots then one is fine.

You can in some cases restore data using snapshots. But for your boot device, the only data that really matters is your config file. Make regular backups of that (to your pool would be good, and there are a number of scripts floating around to do this). That way, if/when your boot device fails, simply install to a new one, upload your saved config file, and you're set.

Even a cheap SSD is orders of magnitude more reliable than a USB stick. It will also perform better, though that's going to have a minimal effect on anything (due to ZFS caching, most of the OS lives in RAM most of the time, so the biggest performance difference will be with installing updates). If you have a spare SATA port (which you should), a SSD will really be a better way for you to go than using a USB stick for a boot device.

I decided I wanted to go with a mini ITX setup instead. The node 804 is just too big for what im looking for. Is this motherboard alright?
