Remove users from AD

So after 2 hours of waiting for all the users to download from the Active Directory server, I'm now seeing that anytime I want to change permissions on a share it take about 25mins for the actual pop-up to appear. I'm going to guess it's because it's loading all several thousand users. Is there a way I can remove all the users that were loaded from AD?
